The Impact of Genetic Polymorphisms of The Transforming Growth Factor B3 Gene on Production Traits and Growth Performance in Broiler Chickens

Abstract

This study was conducted in the Poultry field at the Research Station of the College of Agriculture and Marshes, University of Thi-Qar, from 15/9/2024 to 16/3/2025. The study consisted of two phases: fieldwork and laboratory work. 300 Ross 308 broiler chickens were raised from day-old to 35 days. Birds were numbered with plastic tags and weighed weekly to calculate weight gain and feed conversion efficiency. Weekly body measurements were taken, including bird length, chest circumference, and chest width. Blood samples were collected from the wing vein, and broilers were slaughtered to determine carcass yield and weights of primary and secondary cuts. DNA extraction and genotyping of the TGF-B3 gene were performed at the Marshes Research Center Laboratory, University of Thi-Qar. The aim was to determine the genetic structure of the TGF-B3 gene and study its relationship with some production traits in broiler chickens. PCR products were sent to Macrogen (Korea) for sequencing to determine the nitrogenous base sequence of the studied fragment of the gene. The studied fragment was registered in the genetic bank. Two mutations (321.C>T and 290.A>G) were detected in the second intron of the TGF-B3 gene, which is a non-coding region that does not affect the protein structure. Significant differences (P≤0.05) were found in feed conversion efficiency in the second week for the A>G.290 mutation, with AA and AG genotypes outperforming the GG genotype. No significant differences were observed in body weight, weight gain, bird length, chest circumference, and chest width for both mutations.
